Thin Layer Chromatography (TLC) is an important experimental technique that enables the rapid separation and qualitative analysis of small amounts of substances, and is a more practical and effective method for trace separation and analysis. Thin layer chromatography can be used to separate and extract natural products, including alkaloids, glycosides, sugars and fatty acids. In TLC, the stationary phase is a thin layer of adsorbent material coated on the surface of an inert carrier plate. The sample is spotted onto one end of the thin layer plate and inserted vertically into a sealed chamber containing an organic solvent. The mobile phase moves upward on the plate by capillary forces, and each component of the sample migrates different distances due to its difference in affinity for the stationary and mobile phases. When the solvent reaches the top of the plate, the thin layer plate is removed from the unfolding chamber and dried. The separated components are presented as spots on the plate, from which the specific shift value (Rf) of each component is calculated. Also, the spots can be scraped off and the natural product monomers can be extracted by passing them through the column.

Thin layer chromatography steps

  • Thin-layer plate preparation

Thin layer is prepared by stationary phase and water in a certain ratio. The thin layer is applied to a glass plate and then dried.

  • Spotting

The sample is spotted on the thin layer plate with a spotter, usually as a round spot, and the diameter of the spot and the distance between the spots are the same as those of paper chromatography.

  • Immerse in the spreading agent

Put the sample into the unfolding agent of the unfolding chamber, immerse into the unfolding agent to a depth of 0.5~1.0 cm from the bottom edge of the plate, seal the chamber cover, and then remove the plate to the specified distance, dry, and test according to the regulations under each species.
