Flavonoids Detection

Flavonoids, a class of polyphenolic compounds produced by phenylalanine metabolism, play an important role in physiological processes in plants, such as root growth, flower pollination, pathogenic bacteria and UV stress. Natural flavonoids exist mostly in the form of glycosides and can form a variety of flavonoid glycosides depending on the type, amount, linkage position and linkage mode of the sugars. BOC Sciences used HPLC-MS for absolute quantification of flavonoid monomers.

Flavonoids Detection Methods

Flavonoids can be determined by UV spectrophotometry, thin layer chromatography, high performance liquid chromatography (HPLC), capillary electrophoresis, and liquid chromatography-mass spectrometry (HPLC-MS).

  • UV-visible spectrophotometric method is the most used in the determination of total flavonoids because of its high sensitivity, high accuracy, easy operation and wide range of application.
  • Thin-layer chromatography can be used for rapid separation and qualitative analysis of small amounts of substances, and also for tracking the reaction process.
  • Capillary chromatography has the advantages of low mobile phase and sample volume consumption, low environmental pollution, and cost saving, and is especially suitable for the analysis of trace, multi-component, and complex matrix components.
  • Liquid chromatography (HPLC) is a widely used method for the determination of flavonoids in recent years, which has the advantages of easy pre-treatment and operation, high sensitivity and good stability. HPLC-MS method has the advantages of easy pre-treatment and operation, high sensitivity, good stability, etc. HPLC-MS method can be used to analyze the structure of very small amounts of chemical components in various crude extracts without the presence of controls by adding mass spectrometry to the chromatography.

Sample Requirements

  • Sample preparation
  1. The control group and experimental group need to be set before sampling, and 3 or more replicates are recommended for each group.
  2. Samples need to be taken quickly and as consistent as possible in terms of time.
  3. It is not recommended that customers do the sample extraction, just send us the sample after taking it. If customers complete the extraction step by themselves please note the solvent or method.
  • Storage and transportation

Samples should be placed in appropriate sample tubes with the name of the sample and other relevant information on the tube. It is recommended that all sample tubes be secured in 50 ml centrifuge tubes with caps before shipping, and that the 50 ml tubes be placed in sealed bags.

  • Sample marking requirements

Double marking, send sample tubes and bags with sample numbers clearly written; samples should not be named with simple letters or data, such as A/B/C or 1/2/3, but require samples to be named with letters plus numbers numbering, such as A1, to avoid duplication.

  • Other

The sample technical requirement form must be mailed with the samples. Avoid prolonging the project cycle due to the inability to check the information in time.
